Experiment Sharing: Grinding Mosquitoes Using A Cryo-Grinder

Mosquitoes, though small, carry multiple pathogens and serve as vectors for diseases like dengue, Zika, and malaria. When infected, they transmit these viruses through blood-feeding, posing serious health risks. Monitoring the pathogens mosquitoes carry and preventing their spread is essential.

Before conducting such experiments, mosquitoes need to be ground by a cryo mill. Frozen mosquitoes to a very low temperature, then ground into fine particles. This method efficiently extracts viruses or pathogens, providing samples for disease monitoring and research. After grinding, the samples can be used for virus detection and genetic analysis, helping researchers identify the pathogens carried by mosquitoes, thus improving disease prevention accuracy.

Experiment Purpose:

Objective: Grinding mosquitoes to extract nucleic acids for pathogen detection, a key step in identifying what pathogens mosquitoes carry. This process ensures accurate virus detection and provides reliable data for disease prevention and monitoring.

Materials & Equipment:

Samples: mosquitoes

Equipment: Welso cryo mill

Experiment Steps:

-Place the whole mosquito and reagents in the grinding tube, adding grinding beads.

-Install the grinding tube in the adapter and place it into the mill.

-Turn on the machine, pre-cool for 30 minutes, set to -10°C.

-After pre-cooling, start grinding with the appropriate settings.

-Once grinding is complete, remove the tube for further experiments.

Results:

The mosquito samples after grinding show no tissue residue and are thoroughly ground. The freezer mill's cooling function effectively preserves nucleic acid integrity. The extracted nucleic acids can be used for subsequent testing to accurately identify the pathogens carried by the mosquitoes, providing critical data for disease monitoring and prevention.
