1.Experimental preparation

Prepare the required reagents and instruments: centrifuge, centrifuge tubes (0.5 ml centrifuge tubes, 1.5 ml centrifuge tubes, 2 ml centrifuge tubes, 5 ml centrifuge tubes, 10 ml centrifuge tubes, 15 ml centrifuge tubes, 50ml centrifuge tubes), microscope, phosphate buffer, samples (mixtures containing proteins), etc.

 

2. Centrifugal separation

(1) Add sample into the centrifuge tube: Add sample (a mixture containing the protein) into a clean centrifuge tube. Make sure that the centrifuge tube does not exceed 2/3 of the volume to prevent spillage when centrifugation.

(2) Equilibrate the sample: Place the centrifuge tube in refrigeration and allow the sample to equilibrate at a consistent temperature for a period of time.

3. Select Appropriate Centrifuge Parameters

(1) Choose the appropriate speed and centrifugation time: According to the characteristics of the sample and the separation requirements, select appropriate centrifuge speed and centrifugation time. In general, higher rotational speeds and longer centrifugation times result in better separations.

(2) Set centrifuge parameters: Set the selected rotation speed and centrifugation time on the centrifuge according to experimental requirements

(3) Use a rotor: Install and select the appropriate rotor accordign to centrifuge models to ensure uniform sample distribution and optimal centrifugation.

 

4. Centrifuge centrifugation

(1) Place the tube into the centrifuge: Place the sample into a centrifuge with pre-set parameters.

(2) Start the centrifuge: Close the centrifuge lid and start the centrifuge to start the centrifugation process. Make sure the centrifuge lid is all closed to keep it safe.

(3)Separation process: The centrifuge will centrifuge the sample by rotating at high speed, and the lid cannot be opened during running.

5. Collect the separated substances

(1) After centrifugation, open the centrifuge tube and carefully take out centrifuge tube.

(2) Separate the sediment and supernatant: Aspirate the supernatant and sediment from the centrifuge tube into different centrifuge tubes.

 

6.Proteins Purification

Concentrate proteins: Concentrate the protein of interest from the supernatant using appropriate methods such as centrifugation, ultrafiltration, or aggregation.

(2) Protein analysis: Protein identification and analysis of samples using appropriate protein analysis methods (e.g., SDS-PAGE, Western blot, etc.).

(3) Purification of proteins: As needed, use column chromatography, electrophoresis, and other techniques to further purify the target protein.

 

7. After Experiment

(1) Clean the centrifuge: After completing the experiment, clean the centrifuge to ensure that no contaminants remain.

(2) Data Record and Analysis: Key experimental parameters, results, and observations for protein separation and purification are documented and appropriately analyzed.
